En términos generales, mi trabajo se concentra en el metabolismo del pre-mRNA y los RNAs no codificantes, en particular de los parásitos protozoarios. En el proceso de descubrir el primer cariotipo molecular de Entamoeba histolytica y E. invadens, encontré evidencia de los componentes quitina sintasa y quitosoma de entamoeba, así como el protozoario homólogo Myc. Debido a la falta de evidencia bioquímica, me propuse analizar las funciones distintivas de las proteínas espliceosómicas del parásito. Usando varios enfoques de expresión molecular y expresión in vivo, hemos revelado los mecanismos de las proteínas de empalme constitutivas y centrales que afectan el empalme de los intrones de entamoeba llevando a sitios de empalme canónicos y no canónicos. Mi interés se centra en el impacto de las interacciones RNA-proteína y la estructura secundaria del RNA en el resultado del empalme y la regulación de la expresión génica. También descubrí RNA intrónicos circulares no codificantes de longitud completa (flicRNAs), tanto nucleares como citoplásmicos, y también se caracterizaron los circRNA exónicos utilizando minería de datos in silico. La evidencia sugiere que, mientras que los flicRNA nucleares regulan a la baja su expresión génica parental en cis, se están realizando análisis proteómicos, metabolómicos y de ChIP para revelar las funciones de los restantes RNA circulares no codificantes.
